Domenico was born about 1855 in Rocca Pia, L'Aquila, Abruzzo, Italy, the son of Vincenzo Letterio and Maria Scolastica DeSantis.
His wife is not known. They were married, but the date and place have not been found. Their only known child was Giovanna (1891-1961).
